Abstract

In thepresentstudy,theeffectoforexin-A(ORXA)microinjectionintotheparagiganto- cellularis lateralis(LPGI)onnociceptivebehaviors,usinghot-plateandformalintestsas thermal andchemicalmodelsofpaininrat,wasexamined.Also,wedeterminedwhether the pretreatmentwithSB-334867,aselectiveOX1-receptorantagonist,wouldpreventthe antinociceptiveeffectoforexin-A.ORXA(0.1–100nM/0.5 mL) microinjectedintotheLPGi nucleus, dose-dependentlydecreasedtheformalininducednociceptivebehaviorsandalso produced adose-dependentantinociceptiveeffectinthehot-platetest.Pretreatmentwith a selectiveorexinreceptor1(OX1R)antagonist,SB-334867,alsoinhibitedtheeffectof ORXA onformalininducednociceptivebehaviorswhiletheSB-334867(100 mM) alonehad no effectonformalintest.ThesedatademonstratedthattheORXA-inducedantinocicep- tion informalintestismainlymediatedthroughtheOX1RinLPGiwhichmightplaya potential roleinprocessingthepaininformationassociatedwithdescendingpain modulation.
