Abstract

Isolation and gnotyping of fasciola in Qazvin city Abstract Background: Fascioliasis is one of the most important zoonotic diseases associated with increasing human health problems that occur in different parts of Iran by Fasciola hepatica and Fasciola gigantica. Molecular methods have decisive for the identification of fasciola species. Objective: This study was aimed to detect the speices of fasciola by amplifying the ITS-2 sequence in Qazvin. Materials & Methods: Overall, 30 adult flukes were collected from livestock of Qazvin. DNA extraction and PCR amplification of ITS2 gene region was conducted and phylogenetic tree was constructed. Results: All isolates obtained from cattle and 82.6% of sheep isolates were infected with F. hepatica species whereas 17.4% of sheep isolates were identified as F. gigantica. It was also shown that F. hepatica was the predominant species of the parasite in the region. Conclusion: Both species of Fasciola heptica and F.gigantica were identified in Qazvin, that F. hepatica was the predominant species in the region. Further molecular studies are needed to prove the presence or absence of intermediate fasciola species in the region. Keywords: Fasciola hepatica, Fasciola gigantica, PCR, ITS-2, Qazvin.
